### Lipinski's Rule of Five (Ro5)

## hERG Cardiac Toxicity Risk
Structural alerts: basic nitrogen (pKa 79) + aromatic ring + hydrophobic moiety, LogP > 3.5 + basic amine.
Mitigation: reduce basicity, introduce polar groups, break planarity.
